Validity of Local Positioning System

The aim of this presented thesis is to verify the validation of LPS, which runs on the principle of wave radiations, UWB technology in particular. The testing protocol verifies the precision of system functions in various places and two diferent altitudes of the pitch. The ultimate precision of monitoring was recorded in the middle of the pitch, vanishing to the sides. In a higher altitude, with no surrounding walls, the recordings from the middle of the pitch were nearly similar to the sides of it. The anchor altitudes of the mechanism affect the overall monitored results. In comparison, the accuracy of tag placed approximately in a plane of anchors was lower than tag placed in a vest of an athlete.
Electrical Installation Project for Kindergarten

The work has three main parts. The first part of the thesis introduces the problems of power and data installations to the reader. This includes related legislations, LPS, the methods of feeding the building by electrical power and the electrical wiring in the building, including their execution and protection. In the second part of the thesis are defined the principles of creation of electro design documentation. Mainly the requirements for their content and editing. An important point is the process of creating a design. The last part is practical. It is the elaboration of a complete wiring design of the kindergarten building, which consists of drawings and technical reports.

This bachelor thesis deals with the creation of electrical documentation for a commercial building. It aims to introduce theoretical problems of electrical design in general and the processing of implementation project for a woodworking workshop. The theoretical part of the thesis is mainly devoted to important findings and normative requirements for high-current, low-current distributions, and a draft of external and internal lighting protection. The practical part of the thesis deals with the processing of the implementation project of electrical drawing and text parts for a woodworking workshop. The project documentation includes the preparation of a technical report, the determination of the protocol on external influences, a draft of socket and light circuits, including lighting calculation, complete lighting protection, and risk management, electronic fire system, and a draft of the main and the sub-switchboard of the building.

The theoretical part summarizes the requirements that must be followed according to the relevant legal standards, and discusses the aspects of both power circuits including LPS and lighting systems and low-voltage circuits including fire alarm system. The practical part of the bachelor thesis focuses on the design of project documentation of heavy and low-current circuits, including technical report and budget.

Antioxidant and antiinflammatory effects of bilirubin.

For a long time, bilirubin (BR) has been considered a waste molecule with potential toxic effects especially on the central nervous system. Later, it was found that BR exhibited cytoprotective effects and mildly elevated BR levels showed antioxidant, anti-inflammatory and immunomodulatory properties, however, exact mechanisms of the anti-inflammatory actions of BR have not been fully understood yet. The main aim of this study was to assess the protective effects of BR using experimental in vivo and in vitro models in relation to inflammation and oxidative stress. Partial goal was to establish validated analytical method for determination of BR and lumirubin. Gunn and heterozygous rats were treated with lipopolysaccharide (LPS, 6 mg/kg, IP) or vehicle (saline). After 12 hours, blood and organs were collected for analyses of inflammatory and hepatic injury markers. Primary rat hepatocytes were treated with BR and TNF-α, HepG2 and SH-SY5Y cell lines were treated with BR and chenodeoxycholic acid. LPS-treated Gunn rats had a significantly decreased inflammatory response and hepatic injury compared to LPS- treated normobilirubinemic controls. We found different profile of leukocytes subsets and decreased systemic mRNA expressions and concentrations of IL-6, TNF-α, IL-1β and IL-10 in Gunn rats. Hepatic mRNA...
Usage of formylmethionin derivates in tumor immunotherapy (Model melanoma B16-F10)

Cancerous diseases and alternatives of their therapy belong to one of the most discussed issues of present day. Cancer is one of the most common lifestyle diseases. It is estimated that external factors cause the origin of 75 % of all cancerous diseases. The most influential external factors are hectic lifestyle, obesity, wrong eating habits, smoking, excess of alcohol intake, inappropriate sexual behaviour and negative approach to preventive check-ups and at last the permanently increasing lifespan of the population. The most serious problem is the occurrence of malignant tumors that are diagnosed so late. Therefore, the aggressive therapy is often the choice of treatment. But this kind of therapy often irretrievably damage the health of the patient. Modern medicine tries to find some new ways of cancer therapy. One of them is treatment of tumors by targeted immunotherapy using the naturally occurring matters. The main aim of this bachelor thesis is to describe and verify the possibility of treatment of malignant melanoma by formylmethionin derivates. The bachelor theses consists of two parts. Theoretical part is focused on describing the issue of cancerous diseases, tumor formation and classification. It focuses on tumors of skin, their diagnostics and possibilities of the therapy, especially of the immunotherapy by innate immunity activation. In experimental part of thesis is described the cultivation of melanoma cell line B16-F10 for in vitro experiments and the transplantation of cells into syngeneic mice strain C57BL/6 from Charles River Laboratories. Melanoma cells B16-F10 in the number of 400 000 cells were transplanted to 8 weeks old mice, which had a weight of 18 - 20 grams. The methods of measuring tumor size and comparing tumor temperature versus mouse body temperature is also described. The result of the first experiment concludes, that therapy of B16--F10 tumor by f-MLF-(K)12 motif bound by charge do not work, because it does not cause phagocytic attack. In second experiment it was found out that B16-F10 tumor therapy by 3mM f-MLF alone does not work, because it does not cause tumor reduction without the LPS in combination. In the third experiment f-MLF motif was used. It was anchored by lipophilic chain with stearic acid, it was used alone or in combination with LPS. In this experiment the complete temporary elimination of tumors occurred, but after therapy termination the tumor growth appeared almost immediately. The mouse treated by this compound survive for more than 52 days than control mice. Due to experiments performed in this bachelor thesis was found out, that anchoring of formylmethionin peptides to tumor cell have positive effect to decrease of tumor volume in successful immunotherapy.
